Current Research:

To investigate changes in the tumor microenvironment associated with disease progression and in response to treatment to design better therapies. Currently active research areas include:

  • engineering more efficient "tumor-killing" vectors
  • finding and testing novel prodrug/gene therapy strategies
  • defining more selective oncolytic viruses
  • identifying biomarkers predictive of response to therapy.
  • defining the role of host defense response in vector delivery in tumors.

Techniques:

  • Animals:  analysis of transgenic mice; small animal survival surgery
  • Cell biology:  cell apoptosis assay; immunocytochemistry; immunoprecipitation; mammalian cell transfection; RNA interference; viral infection
  • Imaging:  fluorescence and phase contrast confocal microscopy of live and fixed cells; live-animal imaging techniques; microscopic and autoradiographic image processing and analysis and three-dimensional reconstruction and visualization;
  • Molecular biology:  cloning viral and nonviral vectors; construction of viral and nonviral expression vectors; DNA-protein interactions; ELISA; flow cytometry; FPLC; gene and microRNA arrays; in situ hybridization; Northern, Southern, and Western blotting; quantitative RT-PCR; RNA isolation; site-specific mutagenesis
